Monocytes were untreated or preincubated for 90 minutes with SB203580 (10 μmol/L) and then stimulated with anti-CD11b (5 μg/mL, A), anti-CD11c (5 μg/mL, B), ZZ-CD23 (1 μg/mL, C), or MBP-CD23 (1 μg/mL, D) for the indicated times. Panel E: Dose-dependent inhibitory effect of SB203580 on the production of IL-1β. Human monocytes (150 × 103 cells per well) were pretreated for 90 minutes with SB203580 at various concentrations and then incubated for an additional 16 hours with anti-CD11b (20 μg/mL), anti-CD11c (5 μg/mL), ZZ-CD23 (2 μg/mL), or MBP-CD23 (1 μg/mL). IL-1β production was determined in total cell lysates by ELISA. Data are means from triplicates of 1 experiment representative of 2 others (in each condition the SD was less than 10%).
