Cells were stained for the donor marker (Ly-5.2, allophycocyanin), lineage markers (Cy5-PE), c-kit (PE), Sca-1 (Texas Red), and GFP (fluorescence in the fluorescein isothiocyanate channel). The numbers in parentheses indicate gating of lineage-marker negative-to-low cells according to the numeric scale given on the top of the lineage-marker histogram. Donor-derived, LT-HSC will be present at high purity in the Ly-5.2+, Sca-1+, c-kit+, Lin (0-80) gated population.
