Endothelial activation caused by sickle monocytes is inhibited by anti-TNF-α and IL-1β monoclonal antibodies.
Sickle mononuclear leukocytes were isolated and preincubated for 15 minutes at room temperature with blocking antibodies to TNF-α or IL-1β, individually and in combination (10 μg/mL of each IgG). The leukocyte/antibody mixtures were incubated with HUVEC for 4 hours at 37°C (1.5 mononuclear leukocyte/endothelial cell). HUVEC E-selectin was measured by enzyme immunoassay, and the results are expressed as a percentage of control (media only)-treated HUVEC (100%).
