Superior stimulation of CD8+ T cells by SL-encapsulated antigen.
Mouse BmDC were generated and transferred to new wells on day 6. On day 7, antigens (OVA-SL or sOVA) were added at 10 μg/mL, and DC pulsed with empty SL (based on lipid concentration) or cultured in medium alone were controls. On day 8, cells were harvested, washed, and added in graded doses to 3 × 105 CD8+ T cells from OT-1 mice (OVA TCR-transgenic mice). After 24 hours T-cell proliferation was assayed by thymidine incorporation, and mean log cpm × 10−3 ± SEM of triplicate cultures from a representative experiment are shown.
