Binding of ¹²⁵I-VEGF to polymerized fibrin.

(A) ¹²⁵I-VEGF was added to a solution of 100 μg/mL fibrinogen and then clotted by the addition of 0.5 U/mL thrombin. Bound and unbound VEGF were then separated by vacuum filtration, and non-specific binding (squares) was determined in the presence of 10-fold molar excess of VEGF. Specific binding (triangles) was calculated by subtracting non-specific from total binding (diamonds). Each point represents the mean ± SD of 3 different experiments. (B) Scatchard analysis. The best fit of the data was determined using the Ligand program and indicated the presence of a single binding site.
