Fibrinogen inhibits ADP-evoked cytoskeletal association of p60src.
Dimethyl BAPTA-loaded human platelets were stimulated for 3 minutes with no addition or with ADP (5 μg/mL) in the absence or presence of fibrinogen (1 mg/mL) and then lysed with a Triton X-100 buffer and centrifuged to separate the detergent-insoluble fraction. These samples were subjected to SDS–PAGE and analyzed by Western blotting with the specific anti-p60src antibody GD11, as described in “Materials and methods.” Bands were revealed using chemiluminescence and were quantified by scanning densitometry.
