Effect of platelet-type vWD mutations on vWf-binding with decreasing concentrations of ristocetin.

Transfected cell lines were incubated with increasing concentrations of vWf (0-4 μg/mL) at decreasing concentrations of ristocetin: (A) 0.75, (B) 0.5, and (C) 0.25 mg/mL. Bound vWf was detected using an FITC-conjugated anti-human vWf antibody, followed by flow cytometric analysis. Data are expressed as a ratio of the mean fluorescence of vWf binding to that of AK3 binding. Mean ratio ± SEM from 3 experiments is shown.
