Identification of sHbSR in plasma.

Immunoblotting with the monoclonal GHI/61 antibody of purified membrane-bound HbSR, recombinant sHbSR (extracellular domain expressed in CHO cells), and sHbSR immunoprecipitated (with polyclonal anti-HbSR IgG) from plasma. Because of hyperglycosylation of the sHbSR expressed in CHO cells, the electrophoretic mobility was also compared after deglycosation with PNGase F. (B) Immunoblotting with the monoclonal GHI/61 antibody of sHbSR immunoprecipitated from different plasma samples with different levels of sHbSR as measured by the sandwich enzyme-linked immunosorbent assay (ELISA). Enhanced chemiluminescence was used as the detection system.
