Cytoplasmic localization of FANCA phosphorylation.
(A) Cytoplasmic localization of phosphorylated FANCA and FANCA-PK activities. HSC72/wt-FANCA cells (approximately 3 × 107cells) were metabolically labeled by 32P and fractionated to cytoplasmic and nuclear fractions; then each fraction was subjected to immunoprecipitation with anti-FANCA antibody. In vitro FANCA phosphorylation was examined using each fraction from HSC72/wt-FANCA cells (approximately 3 × 107 cells). The middle panel shows Western blots of FANCA. In parallel experiments, each fraction from the cells (approximately 5 × 105 cells) was subjected to Western blotting with antibodies against c-Abl and β-tubulin, nuclear and cytoplasmic markers, respectively (lower panels).
