Effects of wortmannin and okadaic acid on FANCA phosphorylation.
(A) Wt-FANCA in GM6914 cells was immunoprecipitated and phosphorylated in vitro, in the presence of various concentrations of wortmannin. (B) GM6914/wt-FANCA was metabolically labeled with32P for 2 hours in the presence of various concentrations of wortmannin, then FANCA phosphorylation was examined. (C) GM6914 cells/wt-FANCA was labeled with 32P for 2 hours in the presence of okadaic acid (1 μM) or cyclosporine (10 μg/mL), lysed, and subjected to immunoprecipitation with anti-FANCA antibody.
