(A) Subcellular localization of the indicated RUNX1 proteins was detected by immunofluorescence staining with anti-αB1. A schematic of the RUNX1 protein is shown with the Runt and transactivation domains indicated. The sequence of the NLS is indicated. (B) Double staining for mutant RUNX1 and PEBP2β/CBFβ reveals that the subunits are colocalized in the cytoplasm within speckled dotlike structures for the missense mutants and a diffuse staining pattern for the frameshift and nonsense mutations.
