Reduced ability of sDCs to stimulate T-cell proliferation when cultured with anti–IFN-α/β.
The sDCs (96% CD11c+) isolated from B6 mice were cultured for 18 hours in the presence (circles, dotted lines) or absence (squares, solid lines) of anti–IFN-α/β immunoglobulin, washed, irradiated, and used as APCs for TCR transgenic T cells. (A) Different numbers of sDCs and 105 CD8+ responder T cells purified from 2C TCR transgenic mice were added to wells with (open symbols) or without (solid symbols) antigenic peptide (Ser-Ile-Tyr-Arg-Tyr-Tyr-Gly-Leu at 0.05 nM). (B) Different numbers of sDCs and 105 CD4+ responder T cells purified from DO11.10 TCR transgenic mice were added to wells with (open symbols) or without (solid symbols) antigenic peptide (OVA aa 323 through 336 at 144 nM).
