A representative experiment showing impaired capacity of allogeneic CTLs to lyse H-2d–expressing A20 target cells after depletion of alloreactive T cells to H-2d by AICD (●) in comparison to untreated activated allogeneic control cells (▪). Lysis of A20 was antigen specific because H-2b EL-4 target cells were not recognized by untreated alloreactive CTLs (■) or anti-CD95–treated CTLs (○). CTLs were generated by allogeneic stimulation of purified CD90+ T cells (H-2k) with splenic stimulator cells (H-2d) in the presence of anti-CD95 in bulk culture MLC for 5 days and recovered cells incubated with chromium-labeled A20 or EL-4 target cells for 5 hours to be analyzed for their residual lytic capacity to H-2d.
