Mismatch PCR and restriction enzyme analysis.

(A) DNA prepared from total white blood cells from the patient and purified cellular fractions from the father. Percentages of mutant DNA are derived from radioactive PCR analysis. Cell purity (November only): CD3⁺, greater than 98%, CD14⁺, 85% (by flow cytometry); neutrophils, greater than 97% (400 cell differential).
