Figure 2.
In vivo selection of GFP+ peripheral blood cells in monkey RQ2278 following treatment with TMTX and NBMPR-P. (A) This panel shows flow cytometry analyses for GFP expression in gated peripheral blood cells both prior to and 8 days after the initiation of drug treatment. Lymphocytes, monocytes, and granulocytes were gated on the basis of forward and side scatter characteristics as shown (left panel). Blood from an animal not receiving a transplant was used to set the gates as shown. Baseline proportions of GFP-expressing granulocytes (top row), monocytes (middle row), and lymphocytes (bottom row) are shown in the left panel. Percent GFP+ cells are indicated for each histogram. The right panel shows the proportions of GFP-positive cells detected 8 days after the initiation of the drug treatment course. (B) The percentage of GFP-positive peripheral blood cells measured 4 days before the initiation of the first treatment course and 1, 3, 7, and 8 days after drug treatment are shown. Values for granulocytes (▪), monocytes (▨) and lymphocytes (▦) are shown.
