Similar self-renewal capacity seen for AML1+/– and +/+ LTR-HSCs. (A) 1 × 106 BM cells (Ly5.2+) from AML1+/+ or +/– mice were injected into lethally irradiated recipients. Three to four months after transplantation, BM cells were harvested from animals that had received transplants, donor-derived Ly5.2+ cells were sorted by FACS, and 0.5 to 1 × 106 Ly5.2+ cells were injected into the secondary recipients. This procedure was then repeated sequentially 3 to 4 months after each transplantation. The percentage of mice surviving from 4 sequential cycles is shown (n = 9-10 mice/group). (B) BM cells from secondary recipients were analyzed for multilineage engraftment by FACS.
