Dual role of PAR1. PAR1-dependent signaling by the interdependent procoagulant and anticoagulant proteases thrombin and APC can have opposite effects on endothelial barrier integrity. Barrier protection by APC or low concentrations of thrombin is mediated by sphingosine kinase-1 activity and crossactivation of S1P₁ signaling.
