PPF in BACH1 transgenic mice and their wild-type littermates. The megakaryocyte fractions (enriched by a BSA density gradient) were incubated in IMDM supplemented with 1 × Nutridoma-SP. After 20 hours of incubation, acetylcholine esterase activity was detected before counting the megakaryocytes to determine their PPF. The number of acetylcholine esterase-positive cells displaying the obvious filamentous cell projections was counted and the PPF ratio (number of megakaryocytes with PPF–total megakaryocytes) was calculated.
