RPS19 silencing increase the myeloid-erythroid ratio in mixed liquid culture. (A) Proliferation in mixed expansion medium (X-vivo-15 containing FCS [20%], penicillin-streptomycin [100 U/mL], SCF [20 ng/mL], IL-3 [20 ng/mL], G-CSF [100 ng/mL], Epo [2 U/mL]) decreased in RPS19-silenced cells. Statistically significant reduced cell counts were seen in LV-TH-RPS19-C-transduced cells at all times and in LV-TH-RPS19-B-transduced cells on day 14. (B) As in cells cultured in erythroid expansion medium the GFP+ Glycophorin A+ fraction decreased in RPS19-deficient cells on day 7. In myelo-erythroid medium, however, the fraction of CD13+ and CD4+ cells increased.
