The cover confocal microscopy image shows murine embryonic fibroblasts (MEFs) expressing mutated CALM-AF10 lacking the hydrophobic residues of the CALM-derived nuclear export signal (NES). Mutant CALM-AF10 is detected with an anti-Flag primary antibody and 568-conjugated anti-mouse secondary antibody (red), and nuclei are stained with DAPI (blue). Although CALM-AF10 normally localizes to the cytoplasm, point mutation of the NES prevents nuclear export and results in exclusive nuclear localization of CALM-AF10. The authors determine the CALM-derived NES is necessary and sufficient for CALM-AF10-mediated leukemogenesis, emphasizing a critical role for the nuclear export pathway in these leukemias.
