Asymmetric cell division as a mechanism for hematopoietic stem cell (HSC) self-renewal remains enigmatic. The endocytic gene, Ap2a2, is shown to impact upon HSC self-renewal while also demonstrating asymmetric segregation during real-time videomicroscopy. In these still images taken from a videomicroscopy of a CD150+48− LSK stem cell transduced with a cherry-Ap2a2 fluorescent fusion, Ap2a2 is seen to clearly localize at one polar side during telophase, and then asymmetrically segregate into only one daughter cell after cytokinesis.
